Charles Lister was born on 3 October 1890 in Hawthorn, a suburb of Melbourne, and attended Hawthorn College before going on to study medicine at the University of Melbourne. He served as a medical officer with the Royal Australian Army Medical Corps (RAAMC), reaching the rank of captain. On 18 December 1916, in Holy Trinity Church, Melbourne, Charles Lister married fellow tennis player Meryl Waxman.

Charles and Meryl Lister welcomed a son, John Mervyn Lister, on 7 February 1918. Charles Lister died of influenza on 21 November 1918 while on service in the commune of Wimille in the Pas-de-Calais department, close to the coast in northwest France. He was 27. He was buried in Terlinchtun British Cemetery in Wimille.
